The aim of this randomized, controlled, clinical trial is to compare the clinical efficacy of a desensitizer that contains calcium phosphate and forms hydroxyapatite with a two-step self-etch adhesive (Clearfil SE Bond 2, Kuraray) and a placebo (distilled water) through a long-term period (6 months). Fifty patients between the ages 21 and 64 with a sensitivity score of 6 or higher, according to the Visual Analog Scale (VAS), in at least three teeth were selected from a total of 200 patients. Teethmate Desensitizer, Clearfil SE Bond 2 and placebo (distilled water) were applied randomly to three teeth of each patient. Recalls were done at baseline, 1 week, 1 month, 3 months and 6 months after treatment by applying air blast and the sensitivity scores were determined according to VAS. Replicas were taken from 10 randomly selected patients by using of polyvinyl siloxane, before and 10 minutes after applying Teethmate Desensitizer, Clearfil SE Bond 2 and placebo at control visits. Epoxy resin models were obtained from these replicas. SEM analysis of the obtained models was performed. The VAS scores and SEM images of the same teeth did not correlate. The success of the applied materials was determined according to the patients’ satisfaction. Cochran Q and Dunn pairwise comparison tests were used for statistical analysis of the data (p = 0.05). The efficiency of the materials used in the present study was determined according to the VAS scores. Data were statistically analyzed using Paired t test and repeated measures of ANOVA with Bonferroni correction (p = 0.05). Since the Placebo treatment was only successful in a small number of teeth,the comparison of the three materials could only be done 10 minutes after the treatment. At the next times only Teethmate Desensitizer and Clearfil SE Bond 2 were compared. It was observed that sensitivity decreased significantly (p <0.05) after applying each of the three materials when compared to the condition before the treatment. There was no significant difference between the successes of Teethmate Desensitizer and Clearfil SE Bond 2 treatments at all times (p> 0.05), while placebo treatment was significantly less successful (p <0.05). Successes of treatments were decreased rapidly in both experimental (Teethmate Desensitizer) and control (Clearfil SE Bond 2) groups during the first week after the application. There was no significant difference between Teethmate Desensitizer and Clearfil SE Bond 2 according to VAS scores (p> 0.05) and both materials were more effective than placebo (p <0.05). According to the results of this study calcium phosphate based Teethmate Desensitizer and two-step self-etch adhesive Clearfil SE Bond 2 can be applied to treat hypersensitivity.
