Abstract
Uzun, M., Development of Collagenous Tissue Restorative Phytocosmetics on Skin by Investigation of MMPs Inhibitory Activities, Antioxidant Capacities and Spf Values of Rumex crispus L. Extracts and Sekonder Metabolites, Hacettepe University, Institute of Health Sciences, Pharmacognosy Program, PhD Thesis, Ankara, 2017. The main objective of this thesis is providing effective, high-quality and safe phytocosmetic's material and final product. Inhibitory effects of 15 extracts -with different polarities prepared from the root, leaves and fruits of the plant- and secondary metabolites on MMP-1, MMP-8 and MMP-13 enzymes which cause skin aging and SPF determination and sun protection effects were detected for the first time for a phytocosmetic raw material and formulation that can fight against skin aging. Antioxidant capacities were determined by scavenging effects of extracts against DPPH, NO, ABTS radicals and phosphomolybdate method. Emodin, aloe-emodin, chrysophanol, physcion, emodin-1-O-β-glucopyranoside (first time) , emodin-6-O-β-glucopyranoside (first time), emodin-8-O-β-glucopyranoside, chrysophanol-8-O-β-glucopyranoside and nepodin-8-O-β- glucopyranoside compounds were isolated by various chromatographic methods from Rumex crispus L. (Polygonaceae) roots and leaves and structures were determined by spectroscopic methods. Molecular docking studies on antranoit, flavonoid, tannin and naphthalene skeletons with MMPs enzymes were performed for the first time. Stability tests of high activity extracts were performed for the first time and chemical profiles were determined by HPLC analysis. Extracts and pure compounds were found to have high MMPs inhibitory effect, high SPF, high antioxidant capacity and anti-aging thermosensitive gel formulation was developed from fruit ethanol:water (70:30) extract which has the highest activity and product was obtained.
