Abstract
ABSTRACT Gönülalan, E. M. Stability and Biological Activity Studies on Some Medicinal Cultivated Plants with Economic Priority in Turkey, Hacettepe University, Institute of Health Sciences, Pharmacognosy Program, PhD Thesis, Ankara, 2017. The aim of this study is to perform quality control of Silybum marianum L., Achillea millefolium L., A. filipendulina L., Echinacea purpurea (L.) Moench, Mentha piperita L., Salvia officinalis L., Nigella sativa L., N. damascena L. and their finished products by HPLC, to prove the phytocosmetic effects of 4 extract prepared from Achillea millefolium L., A. filipendulina L., Mentha piperita L., Salvia officinalis L., by means of the MMP-1, 8, 13 inhibitory effects, radical scavenging (DPPH, NO, ABTS), antioxidant (FRAP, CUPRAC and total antioxidant) capacities and sun protection factor (SPF) assays, in addition, to perform metabolomics analysis with GC-MS and to interpret the relationship with the activity. MMP 1,8,13 inhibitory effects, SPF determination and antioxidant potential assays were performed simultaneously for the first time. Methanolic extract of Mentha piperita showed the best activity in all tests except SPF. Phytoequivalence studies of herbal medicines and food supplements used for the same purpose but licensed by different authorities were performed by comparing HPLC-chromatographic fingerprint profiles. Many of products were found not to carry desired quantity of chemical markers. The levels of metabolites can be considered as the response of biological systems. A. millefolium, A. filipendulina, M. piperita, S. officinalis showed similar metabolic profiles. The evaluation of all metabolites instead of a single metabolite contributed to our knowledge of synergism or antagonism. Therefore, we compared the whole metabolomic profile of different species and MMP inhibitory activity in order to find metabolites that have negative or positive activity. 67 positive correlations were found between metabolit levels and MMP inhibitory activity (r > 0.70) however 94 metabolites were found to posess negative correlations.
